چکیده

Draft genome sequences for Schistosoma japonicum and Schistosoma mansoni are now available. The schistosome genome encodes approximately 13,000 protein encoding genes for which the function of only a small minority is understood. The new genes represent potential intervention targets. Molecular tools are needed to determine the importance of these new genes. There is a role for transgenesis in functional genomics of the new schistosome gene sequences, both in gainand loss-of-function approaches such as insertional mutagenesis screens and vector-based RNA interference. This laboratory symposium focuses on the development of approaches, systems, and tools to address the problem of establishing transgenic schistosomes which, in turn, can be applied to fundamental questions of schistosome physiology, the host–parasite relationship, and to developing new interventions.

Paving the way for transgenic schistosomes.

In parasitological research, significant progress has been made with respect to genomics and transcriptomics but transgenic systems for functional gene analyses are mainly restricted to the protozoan field. Gene insertion and knockout strategies can be applied to parasitic protozoa as well as gene silencing by RNA interference (RNAi). By contrast, research on parasitic helminthes still lags beh...

Germline Transgenesis and Insertional Mutagenesis in Schistosoma mansoni Mediated by Murine Leukemia Virus

Functional studies will facilitate characterization of role and essentiality of newly available genome sequences of the human schistosomes, Schistosoma mansoni, S. japonicum and S. haematobium. To develop transgenesis as a functional approach for these pathogens, we previously demonstrated that pseudotyped murine leukemia virus (MLV) can transduce schistosomes leading to chromosomal integration...

Establishing a new animal model for muscle regeneration studies

Skeletal muscle injuries are one of the most common problems in the worldwide which impose a substantial financial burden to the health care system.  Accordingly, it widely accepted that muscle regeneration is a promising approach that can be used to treat muscle injury patients. However, the underlying mechanisms of muscle regeneration have yet to be elucidated. The muscle structure and muscle...

Epigenetics in Schistosomes: What We Know and What We Need Know

Schistosomes are metazoan parasites and can cause schistosomiasis. Epigenetic modifications include DNA methylation, histone modifications and non-coding RNAs. Some enzymes involved in epigenetic modification and microRNA processes have been developed as drugs to treat the disease. Compared with humans and vertebrates, an in-depth understanding of epigenetic modifications in schistosomes is sta...

Diversification, dioecy and dimorphism in schistosomes.

In addition to causing one of the great neglected diseases of the world, schistosomes have unusual biological features that further command attention, including their habit of living in the blood of their hosts and the presence of separate, dimorphic sexes. Phylogenetic studies including a more complete sampling of pivotal and rare schistosomes and their relatives, provide an improved framework...
